In addition to apoA-I, the assembly of HDL particles involves the interaction of lipid-free apoB-100 with the phospholipid and cholesterol components of the nascent lipoprotein particles. To define the role of apoB-100 in the formation of HDL particles, recombinant apoB-100 (rApoB-100) was expressed in insect cells and purified to homogeneity. The addition of rApoB-100 to model lipid solutions containing apoA-I, phospholipids, and cholesterol resulted in HDL-like particles containing apoA-I and lipid. When rApoB-100 was added to apoA-I-containing solutions, only a small amount of rApoB-100-containing HDL particles were formed. Similar results were obtained when apoA-I was added to rApoB-100-containing solutions. In the absence of apoA-I, rApoB-100 formed HDL-like particles with lipid but lacked apoA-I. When model apolipoprotein mixtures were used in which either apoA-I was replaced with apoA-IV or apoA-I, apoA-IV or apoA-I, respectively, were added to mixtures containing rApoB-100 and lipid, then rApoB-100-containing HDL particles were formed. These results indicate that both apoA-I and apoA-IV are necessary to form rApoB-100-containing HDL particles in the absence of lipid.
